چکیده

One of the most recent trends in skin irritation safety testing of personal care and cosmetic products, discussed in detail in this review, is the use of reconstructed human epidermis models. Some of the commercially-available models have undergone a series of validation trials, which successfully advanced them as valuable tools for industry use. On a broader perspective, the 3D models are now part of the integrated decision-tree testing strategies used in regulatory settings for skin irritation and/or corrosion registration and labeling of newly designed formulations. Furthermore, the reconstructed human epidermis models are also widely used by industry for product development, safety testing, and ranking of personal care and cosmetic products’ irritation potential. Although the specific protocols used for meeting either legislation-driven regulatory safety testing or nonregulatory, industrial safety screening may differ considerably, the in vitro tissue systems and approaches are aligned to meet the newest legislation or corporate policies prohibiting the use of animals for safety testing.

Alternatives to In Vivo Draize Rabbit Eye and Skin Irritation Tests with a Focus on 3D Reconstructed Human Cornea-Like Epithelium and Epidermis Models

Human eyes and skin are frequently exposed to chemicals accidentally or on purpose due to their external location. Therefore, chemicals are required to undergo the evaluation of the ocular and dermal irritancy for their safe handling and use before release into the market. Draize rabbit eye and skin irritation test developed in 1944, has been a gold standard test which was enlisted as OECD TG 4...

OECD GUIDELINE FOR THE TESTING OF CHEMICALS DRAFT PROPOSAL FOR A NEW GUIDELINE In Vitro Skin Irritation: Reconstructed Human Epidermis (RhE) Test Method

INTRODUCTION 1. Skin irritation refers to the production of reversible damage to the skin following the application of a test chemical for up to 4 hours [as defined by the United Nations (UN) Globally Harmonized System of Classification and Labelling of Chemicals (GHS)](1). This Test Guideline provides an in vitro procedure that may be used for the hazard identification of irritant chemicals (s...

[Development of alternative to animal experiment in evaluation of skin irritation caused by alcohol-based hand rubs].

Alcohol-based hand rubs are widely used for infection control in clinical practice. However, it is known that frequent use of the alcohol-based hand rubs may cause skin irritation. To predict the skin irritation in human, animal experiments are quite useful. Especially, the Draize Test using rabbits is suitable for this purpose because their skin is highly sensitive. On the other hand, the deve...

Dermal xenobiotic metabolism: a comparison between native human skin, four in vitro skin test systems and a liver system.

The xenobiotic metabolism of 4 in vitro human skin test systems (2D and 3D) was compared with that of the native human skin samples from which the skin test systems had been produced. In total 3 skin samples were investigated, each from a different donor to exclude variability due to gender, donor or tissue supplier. In addition, the skin cultures were compared with a surrogate of the liver. Ba...

Characterization and comparison of reconstructed skin models: morphological and immunohistochemical evaluation.

Reconstructed human skin equivalents are currently being investigated as in vitro models for the prediction of human skin toxicity and irritation responses. Three different industrial reconstructed skin models (EpiDerm, Episkin and SkinEthic) and one in-house equivalent were characterized and compared using light microscopy, immunohistochemistry and reduction of (3-[4,5-dimethylthiazole-2-yl]-2...
